Vesicare 10mg overview
The main component of the medicine is solifenacin succinate, which is widely used to reduce the tone of smooth muscles of the urinary tract. It is the newest medicine for the treatment of overactive bladder. In contradistinction to other urinary antispasmodics, Vesicare 10mg is used only once a day.
Indications
The medicine is intended for patients with urinary frequency, urinary incontinence, uriesthesia, nocturia, which are typical for patients with overactive bladder.
How does Vesicare 10mg work?
Vesicare 10mg is a specific competitive inhibitor of muscarinic receptors that has antispasmodic and anticholinergic effect.
After an oral administration, Cmax is reached after 3-5 hours. The absolute bioavailability is 90% and it is independent of food intake. The maximum effect of the medicine can be noticed after 4 weeks. The medicine is extensively metabolized in the liver. After 26 days 70% of the 10 mg dose (1 coated tablet) of the medicine is excreted with urine and about 23% is excreted with feces.
Contraindications and precautions
Do not use Vesicare 10mg if you are hypersensitive to any component of the medicine, if you have urinary retention, myasthenia, neuropathy, moderate or severe renal or hepatic failure, angle-closure glaucoma, galactosemia, severe gastrointestinal disease or if you are on dialysis treatment. Do not use with the active inhibitors of CYP3A4.
Before starting treatment your doctor must exclude urinary tract infection. If you want to achieve the desired effect, the drug should be used for more than 12 months.
Vesicare 10mg is not recommended for use during lactation or if you are pregnant. Use with caution in patients, whose work requires quick physical and mental reactions. The medicine can reduce the effect of drugs stimulating the motility of the gastrointestinal tract.
Adverse effects and the dosage
Possible side effects are associated to the anticholinergic effects of solifenacin. As a rule, they are lowly or moderately expressed and they are dependent on the dosage. The most frequently reported side effect of Vesicare 10mg is dry mouth. It has been observed in 22% of patients who took 10 mg of the medicine (1 pill) per day. The severity of this side effect was mild and did not require discontinuation of the therapy. Less common side effects include sickness, dyspepsia, taste disturbance, stomach-ache, fatigue, urinary tract infection, constipation, leg swelling, strangury, xerosis. Allergic reactions are extremely rare. However, if you have anaphylaxis, you should immediately seek medical help. If you have any adverse effects, you should tell your doctor.
The dosage is established by your doctor. The usual dose is 5 mg (0,5 coated tablet) per day and it is independent of food intake. If necessary, the dose is increased to 10 mg per day. The coated tablets should be taken with a glass of water.
An overdose of solifenacin succinate can potentially lead to severe anticholinergic effects. In case of overdose patients should take the activated carbon.
